Effect of type 2 protease activated receptor (PAR-2) activation in the osteogenic activity of the periodontal ligament stem cells

Abstract

Protease-activated receptor-2 (PAR-2) is associated with the pathogenesis of chronic inflammatory diseases including periodontitis and PAR-2 activation plays a relevant role in the inflammatory process. The aim of the present study was to evaluate the effect of PAR-2 activation on the osteogenic activity of human periodontal ligament stem cells (PDLSCs). PDLSCs obtained from 3 subjects were treated with a control medium or with an osteogenic medium for 2, 7, 14, and 21 days. Cell proliferation, alkaline phosphatase activity (ALP), gene (qPCR), and protein expression (ELISA assay) of osteogenic factors and Calcium deposits, calcium concentration (supernatant) were assessed in the presence of trypsin (0,1U/ml), PAR-2 specific agonist peptide (100nM), PAR-2 antagonist peptide (100nM), and PAR-2 control peptide. The activation of PAR-2 led to alteration on the cell proliferation, a decrease in the formation of calcium deposits (p<0.05), in calcium concentration (p<0.05) and ALP activity (p<0.05). Further, qPCR and ELISA assay showed that activation of PAR-2 may increase gene and protein expression of RANKL (p<0.05), and decrease the gene and protein expression of OPG (p<0.05). The results of the present study demonstrate that PAR-2 activation increases proliferation decrease the osteogenic activity of PDLSCs, indicating that PAR-2 may be an important target to be considered in the use of periodontal ligament mesenchymal cells in regenerative procedures in periodontology.
