Abstract

The activation of the protease activation receptor type 1 by thrombin may play an important role in homeostasis and repair of the periodontal tissues. A recent study from our group demonstrated an increased PAR1 expression in gingival fluid cells in patients with chronic periodontitis after periodontal treatment, and the PAR1 expression was associated with a decreased expression of inflammatory biomarkers. In addition, in vitro studies have shown that the activation of PAR1 by thrombin induces the synthesis of osteoprotegerin (OPG) in periodontal ligament cells, leads to a dose-dependent increase proliferation of cells isolated from bone marrow stroma of rats and induces the expression of growth factors in osteoblasts. Therefore, studies suggests an important role of PAR1 in the periodontal tissue repair. In this study, we hypothesized that activation of PAR1 in the periodontal ligament cells can lead to increased osteogenic activity. Thus, the main objective of this study is to evaluate the effect of PAR1 activation by thrombin on the osteogenic differentiation of ligament cells. Furthermore, will be evaluated the effects of the PAR1 activation on gene and protein expression of alkaline phosphatase (ALP), osteopontin, osteocalcin, RANKL, OPG and runt-related transcription factor-2 (Runx2).
