Abstract

One of the critical steps in the production of second generation ethanol from sugarcane bagasse is the production of enzymes efficiently and inexpensively. In this context, the objective of this project was to study ways to increase the activity of the glycohydrolases produced by Trichoderma harzianu P49P11 by optimization of the culture medium and by classical genetic improvement, , as well as to develop the necessary methodologies to reach this objective. A methodology was developed to optimize small-scale culture media using stirred flasks with pH controlled by potassium biftalate buffer. This methodology allowed several studies that helped to better understand the system of induction of the enzymes by the different culture media studied. It was also developed a methodology to measure the activity of glycohydrolases using sugar cane bagasse as substrate of a hydrolysis carried out in mini scale at 50ºC, and the release of reducing sugars in 2h of reaction was used as an estimate of the activity of the enzymes. In order to obtain the mutants used throughout the work, five cumulative mutations were performed using the EMS chemical reagent. At the end of the genetic improvement process, strain 5PCE15 was selected, which presented advantageous characteristics for the process, such as not producing foam in its culture environment and presenting a higher production of glycohydrolases in a medium containing 2 % glucose (AU)
