Abstract

Osteoarthritis is the main disease affecting joint cartilage and the most common joint disease in the world that causes major disability. The present study aims to evaluate different types of osteoarthritis treatments in the medial femoral condyle of Wistar rats. The animals will be treated with diacerein (50mg / kg), glucosamine (500mg / kg) / chondroitin conjugate (400mg / kg) and human deciduous dental pulp stem cells. Twenty-five male Wistar rats were distributed in 5 groups with five animals (n = 5): control group, negative group, diacerein-treated arthritis group, glycosamine/chondroitin conjugated arthritis group and deciduous dental pulp stem cells treated group. Arthritis will be induced by joint infiltration with zymosan (1mg) in the left knee of the animals. After 14 days, the animals will be treated for sixty continuous days with the drugs diacerein and glycosamine/chondroitin conjugate by gavage. The deciduous dental pulp stem cells group will receive single intra-articular application of 8x105 cells in 4mL of saline solution. After the sixty-day period, all animals will be euthanized using intraperitoneal injection with excess of Ketamine and Xylazine (180mg and 20 mg / kg body weight). The medial femoral condyles will be removed, fixed, decalcified for light microscopy protocol and immunohistochemistry. Morphometry, stereology and expression of Sox5, Sox6, Ihh, MMp8, MMp13 and Collagen 2 will be analyzed. The results will be expressed as means, medians and p values less than 0,05 will be considered significant after ANOVA and Tukey Post Hoc multiple comparison test. (AU)
