SARS-CoV-2 glycosylation: a blueprint structural insight for understanding COVID-19 pathogenesis

Abstract

Severe acute respiratory syndrome-related coronavirus 2 (SARS-CoV-2) is an emerging human virus from the Coronaviridae family. SARS-CoV-2 is the etiological agent of novel pneumonia called COVID-19, coronavirus disease 2019. Due to the rapid increase in the number of cases that have affected countries, WHO declared COVID-19 outbreak an ongoing pandemic. As of April 1st, there are 750,890 confirmed cases and 36,405 deaths globally. Understanding host-virus dynamics is critical for development of prophylactic, diagnostic and therapeutic strategies and epidemiological control of COVID-19. Many features are yet to be understood on SARS-CoV-2 behavior and host response during infection, e.g. the molecular and cellular traits of severe, mild and asymptomatic infections, providing higher understanding of the pathogenesis. In this project we propose the application of proteome-wide spatial and temporal approach of SARS-CoV-2:host interaction, to be addressed in three interconnected fronts: (1) Assessment of activated signaling pathways by systematic quantitative analysis of temporal dynamics in protein and glycosylation changes during SARS-CoV-2 infection of human cells; (2) characterization of proteome and PTMs diversity of SARS-CoV-2; and (3) development of an early diagnostic and prognostic platform based on the proteomic profile of sera from patients with different clinical characteristics. The project proposed will contribute to understand the viral pathogenesis and prioritize biomarkers for rapid diagnostic tests. (AU)
