The oleaginous complex of annatto seed (Bixa orellana) for application in weight control and comorbidities: product development, determination of efficacy and safety

Abstract

The project will be carried out by Rubian Xtract Serviços Ltda., a company in pre-market of extracts for applications in the nutraceuticals, cosmetics and food areas. The objective of the work is to develop products from oleaginous complex extracted from annatto seeds (Bixa orellana) by supercritical fluid, to evaluate the efficacy and safety of products obtained as nutraceuticals for application in body weight control and comorbidities. The extraction will be carried out using a clean technique using supercritical carbon dioxide with additional fractionation steps to allow techniques for the standardization of the bioactive by means of the dosage and mixtures of the main compounds of the complex, geranilgeraniol and tocotrienol. The complex will be tested for safety and toxicity through the tests: AMES test and micronucleus test to verify genotoxicity / mutagenicity effects, and subchronic toxicity test to assess possible toxic effects. Two encapsulated products will be developed from the oleaginous complex: soft gel oil and dry powder emulsion. The products developed will be evaluated for stability and efficacy in parameters such as weight gain, lipid profile, insulin resistance, as well as its associated variables: energy expenditure, inflammation markers (protein C e IL-6), anti-inflammatory markers (adiponectin), glucose and insulin response (hyperinsulinemic euglycemic clamp) and factors linked to satiety (GLP-1, ghrelin and peptide YY) in translational human assay. Considering the preliminary results obtained in PIPE I, with emphasis on the effect of crude oil extract on weight reduction, improvement of glycemic response, serum lipid profile and antioxidant defenses, it is expected to develop products from the oilseed complex of urucum obtained by selective supercritical extraction, which has the differential of maintaining natural synergy of the tocotrienol and geranilgeraniol compounds, as originally found in the seeds, with proven safety, stability and biological performance. In this way it can be used with superior efficacy and safety performance inside the ingredients functional food and nutraceutical markets, being the first tocotrienol-geranilgeraniol fitocomplex sourced from Urucum seeds.
