A novel bioglass of the SiO2 - Na2O - CaO - P2O5 system, developed from marine sponge biosilica, for use in bone tissue engineering: from conception to pre-clinical experimental application

Abstract

The rich biological diversity of marine ecosystems makes them abundant in bioactive compounds favorable to the development of innovative biotechnological products. Several studies have shown, for example, that the biosilica extracted from marine sponges has osteogenic properties and our group have recently demonstrated that it may even have superior osteoinductive effects than those of the gold standard bioactive material, the Bioglass®45S5. In this context, this proposal aims to take advantage of the huge potential of Brazilian marine biodiversity for the development of a new bioglass from the SiO2 - Na2O - CaO - P2O5 system, using biosilica from marine sponges. From its conception to the experimental application, there will be 3 stages: (1) the development and the physical-chemical characterization of the particulate biomaterial and of 3D-printed scaffolds; (2) in vitro studies involving indirect (extracts) and direct (cell-scaffold contact) tests on osteoblast precursor cells and covering various analyzes, from the cytogenotoxicity of the biomaterial to its effects on cell metabolism and function; (3) in vivo studies with the surgical implantation of the 3D printed scaffolds in bone defects in rats. Thus, the results of these studies will provide data on the processing and the preclinical osteogenic potential of a new bioglass, which will be valuable for the development of highly effective biomaterials, in terms of quality and accessibility to the citizen, for the treatment of bone tissue disorders from the sustainable use of marine biodiversity of a planet called "blue".
