Abstract

Cardiovascular diseases are the main responsible for mortality in the western population, of which 5% have peripheral arterial disease (PAD). With the worsening of PAD in symptomatic patients without clinical or technical conditions for revascularization of the affected limb, approximately 25% end up evolving to lower limb amputation. Recent researches have shown a promising future in regenerative and tissue replacement therapy involving stem cells and cellular engineering, with clinical application in several diseases, including those of cardiovascular origin. Objective: Promote the development of tissue engineering techniques in blood vessels from an animal model, using scaffolds produced by decellularization of the inferior vena cava, promoting their cell repopulation with autologous mesenchymal stem cells, promoting cell differentiation in vitro, and implanting them in the abdominal aorta to test their functionality in vivo. Methods: 40 female rabbits will be used for all experiments. Inferior vena cava will be obtained from 20 of these rabbits for the production of scaffolds by the standardized process of decellularization with 1% sodium dodecyl sulfate (SDS) and allogeneic vein groups in natura. To obtain mesenchymal stem cells derived from adipose acid, 2g interscapular fat fragments will be collected by aseptic surgical technique and expanded after digestion enzymatic (autologous for the in vivo experiment). There will be two groups in the in vivo test, where an interposition of the vena cava, allogeneic in natura in the abdominal aorta (n=10) will be performed (Group 1 allogeneic vena cava) compared to the group that will receive an interposition of the new vessel produced in the abdominal aorta ( n=10) (Group 2 New blood vessels with autologous MSC). Product analyzes will be carried out in two stages, basically by histomorphological, immunohistochemical, and immunofluorescence analysis (AU)
