In vitro comparative study of the organic and inorganic component of sclerotic and healthy dentin from bovine versus human

Abstract

The sclerotic dentin has been commonly found in elderly patients. This kind of substrate shows typical characteristics of sclerosis: a brownish, smooth and shiny surface (glassy appearance). Several in vitro and in vivo studies have reported micromorphology and bond strength differences between sclerotic and healthy dentin. Bovine dentin has been used in research for replacing human dentin for ethical reasons, difficulties for obtaining human teeth and their storage data. However, there are scarce reports in the literature comparing on the use of human and bovine dentin and a few reports using bovine substrate with characteristics of sclerosis. The aim of this study is to compare the organic and inorganic components of sclerotic and healthy dentin from bovine and human. This would be of importance for futures researches for testing dental materials especially those applied in adhesion procedures. Thirty human and 30 bovine teeth will be distributed into 4 experimental groups, as follows: human healthy (n=15); human sclerotic (n=15), bovine healthy (n=15) and bovine sclerotic (n=15). Morphological analysis of the dentin surface topography and nano-hardness will be achieved using Atomic Force Microscopy (AFM) (n=5 per group). The quantitative evaluation (n=5 per group) of the inorganic components will be obtained by Energy Dispersive X-ray analysis (EDX) using Scanning Electron Microscopy (SEM). The organic components will be analyzed through Transmission Electron Microscopy (TEM) in order to verify the morphology and distribution of collagen fibers (n=5 per group).
