Abstract

O objetivo deste estudo será avaliar as alterações clínicas e microbiológicas promovidas pela terapia de raspagem e alisamento radicular (RAR) associada à administração sistêmica do metronidazol (400mg) e amoxicilina (500mg). Serão selecionados 30 indivíduos com doença periodontal agressiva generalizada que apresentem no mínimo 12 dentes com pelo menos 1 sítio com profundidade de sondagem (PS) e nível clínico de inserção =5mm. Os indivíduos serão divididos em dois grupos terapêuticos: Controle - RAR + medicação placebo (MP) e Teste - RAR + metronidazol (400mg) 3 vezes ao dia durante 14 dias (MTZ) + amoxicilina (500mg) 3x ao dia durante 14 dias. A terapia básica de raspagem (RAR) será realizada em 14 dias. Os indivíduos dos 2 grupos experimentais (controle e teste) serão orientados a realizar bochechos com clorexídina 0,12% 2x/dia. O controle químico do biofilme supragengival será iniciado em conjunto com a terapia de RAR e deverá prosseguir por um período de 45 dias pós-terapia. As avaliações clínicas serão realizadas no exame inicial e repetidas após 45, 90 e 180 dias do término da RAR. Amostras de biofilme subgengival serão coletadas de 9 sítios (rasos, moderados e profundos) em cada um dos voluntários e os níveis de 40 espécies bacterianas serão avaliados pela técnica do Checkerboard DNA-DNA Hybridization e a presença de cepas leucotóxicas de A. actinomycetemcomitans pela reação em cadeia da polimerase. A avaliação microbiológica será realizada na consulta inicial, 45, 90 e 180 dias pós-terapia. Diferenças nos parâmetros clínicos ou microbiológicos entre os 2 grupos, em cada tempo experimental, serão avaliadas por meio do teste teste U de Mann-Whitney. Diferenças dentro de cada grupo, entre os tempos experimentais serão avaliadas utilizando-se os testes Friedman e Wilcoxon. A significância estatística será estabelecida em 5%. (AU)
