Abstract

A toxoplamose, uma doença resultante da infecção por Toxoplasma gondii, manifesta-se clinicamente nas formas ocular, cerebral e congênita. O T. gondii utiliza como rota de infecção e invasão, celulas e tecidos que expressam os antígenos dos sistemas histo-sanguíneos ABO (ABO), Secretor (FUT2), Lewis (FUT3) e Duffy (FY). Estes processos biológicos aparentemente independentes podem estar relacionados entre si e influenciar a suscetibilidade à toxoplasmose ocular. O objetivo geral deste projeto é testar a hipótese de que os antígenos dos sistemas histo-sanguíneos ABO, Lewis, Secretor e Duffy estão associados à toxoplasmose ocular. Serão selecionados pelo menos 200 pacientes caucasianos e não caucasianos, consecutivos, atendidos no Ambulatório de Oftalmologia do Hospital de Base da Fundação Faculdade Regional de Medicina de São José do Rio Preto. Os pacientes serão divididos em dois grupos de pelo menos 100 pacientes cada, de acordo com a presença (G-1) ou ausência de lesões exsudativas e/ou cicatriz corioretiniana (G2). Serão colhidas amostras de sangue total e soro para a identificação dos fenótipos e genótipos ABO, Secretor, Lewis e Duffy por reação de hemaglutinação e PCR (PCR-RFLP e /ou PCR-SSP) quando for o caso. A identificação de anticorpos anti-T. gondii será feita com o uso dos métodos ELISA e Imunofluorescência indireta (IFI). Uma ficha de dados epidemológicos será preenchida para cada paciente. Os resultados de ambos os grupos serão comparados com o uso dos métodos qui-quadrado e/ou teste exato de Fisher, com nível de significãncia igual a 5%. (AU)
