Evaluations of aroeira"s extract influence in adhesion and proliferate of human fibroblast and MMP-2 and MMP-9 expression in vitro

Abstract

Brazil displays a savannah's plant biologic diversity that facilitates the study for researchers in different areas. The aroeira (Astronium urundeuva) is one of these plants that have been studying not only in medicine as in dentistry too, with anti-inflammatory and antimicrobial influence potential, farther favor the repair process. The research has the main purpose realize an aroeira's characterization and available the influence of different concentration's aroeira hidroalcoolic extract in human's fibroblast (FGH lineage) adhesion and proliferate, farther the in vitro matrix metalloproteinase 2 and 9 expression (MMP-2 and MMP-9). Aroeira's characterization will be done for partition process and analitic HPLC-"Fingerprint. The cells will be plated 3x10³ well-cells in 10 plates of 24 wells. The media will be substitute to media Eagle modify by Dullbecco complement for serum bovine fetus 10% in aroeira aqueous extract from different concentration (pure extract; 1:10; 1:100; 1:1000; 1:10000) that will be prepared. Each plate will be analyzed in an experimental time that will be 24, 48, 72, 96 and 120 hours after the medium condition addition. After each experimental time the medium containing the aroeira's aqueous extract will be removed, the cells will be washed with PBSA and distributed analyses different realization. The sample collected will be available like cell viability for environment by cells mitochondrial activity test been realized by the MTT's reduction method (brometo 3-(4,5-dimetiltiazol-2-yl)-2,5-difeniltetrazólio), neutral red catcher, cell's coloration violet crystal methods. The MMP-2 and MMP-9 protein analyses will be done by Western Blot, and your activity for zimography. The MMP-2 and MMP-9 transcription will be quantificated for Real Time PCR. The values will be available by ANOVA's method, complemented by Dunn's test when submit to normal or abnormal distribution, respective. The significant level used will be 5% (p<0,05). (AU)
