Study of the aqueous extract of leaves of Qualea grandiflora: phytochemical evaluation and effects on the expression of proteins involved in inflammatory processes

Abstract

The therapeutic use of medicinal plants for prevention and maintaining health is growing daily, and this increase may be related due to existing a natural product that does not provide harm beliefs. However, for proper use of medicinal plants is vital knowledge of its active components and their toxic and therapeutic potential. Some applications in folk medicine for leaves Qualea grandiflora, are related to diseases of inflammatory origin. Some molecules involved in inflammatory processes, both in tissue destruction as in the repair process, are called matrix metalloproteinases (MMPs). Thus the purpose of this study is to chemically characterize the components present in the alcoholic extract of leaves of Q. grandiflora and evaluate the influence of this extract in fibroblasts and pre-osteoblasts, focusing on activity and regulation of MMPs and their inhibitors (TIMP). The characterization of the extract of Q. grandiflora through chromatographic screening process consisting in collecting the healthy leaves, dried and pulverized percolation in 70% ethanol, solvent partition will be held . The characterization of the extracts is performed by identifying the compounds present therein HPLC -PAD -MS; addition, quantification of chemical markers selected by HPLC -PAD is performed. For cytotoxicity tests fibroblasts and pre-osteoblasts are grown in culture medium with different concentrations (crude extract; 1:10, 1:100, 1:1000, 1:10,000) of leaf extracts of Q. grandiflora. After periods of 24, 48, 72 and 96 hours after treatment with different concentrations of the extract is measured by the cell viability test of the reduction of MTT (3 - (4,5 -dimethylthiazol- 2-yl) -2,5 - diphenyltetrazolium bromide) and incorporção crystal violet. A portion of the collected samples will be used for analysis of MMP-2 and 9, which will be performed by zymography, and the expression of TIMP-2 protein is made by the Western blot method. We will perform RT-PCR to quantify the transcripts (mRNA) of MMP-2 and 9 and TIMP-2. The values will be analyzed by the Kruskal-Wallis complemented by Dunn's test when obey the normal distribution and non-normal, respectively ANOVA complemented by Tukey test.
