Abstract

Phytotherapy, the study of the use of medicinal plants and its products in disease treatment and healing processes, it's an age-old practice. However, studies involving particular species showing the evidence of its beneficial effects have only been featured in recent years. Recently, according to studies, the Qualea grandiflora species from Vochysiaceae family has been recognized for its medicinal attributes, showing, antioxidant, analgesic and gastric protection properties, among others. Thus, the Qualea grandiflora has shown potential against many pathological issues, and the understanding of its mechanisms of action is crucial. This study aims to evaluate in vitro cytotoxicity of Qualea grandiflora Mart. leaf extract in MC3T3 cell lineages (mouse pre-osteoblasts) and NIH/3T3 (mouse fibroblasts) and some proteins that may be direct or indirect targeted by the extract. To run the assays of cell viability, matrix metallopeptidase 14 (MMP-14) and hypoxia-inducible factors, different concentrations of hydro-alcoholic extract of Qualea grandiflora Mart. will be administered for 24, 48, 72 and 96 hours. After each period, the cell viability will be evaluated through the MTT reduction method and the MMP-14 and the HIF-1alpha expression analysis will be performed using immunofluorescence techniques. Statistical data are presented as mean and Standard Deviation (SD). Analyzes among three or more experimental groups will be submitted to one-way ANOVA test following by Tukey test and Kruskal-Wallis test following by the Dunn test, when obey the normal distribution or non-normal, respectively. Analysis between only two groups will be evaluated by the T test and possible correlations by linear regression or Pearson test. In studying the Q.grandiflora hopefully result: 1) producing scientific knowledge about: a) the processes and molecules that can be modulated by plant extract in both cell lines; b) possible modulation in the activity of MMP-14 and HIF-1, hence with potential use in inflammatory conditions that have tissue destruction.
