Diagnosis of visceral leishmaniasis in peripheral blood samples using conventional microbiologic and kDNA- based techniques

Abstract

Although known as a rural endemic, the American Visceral Leishmaniasis (LVA) presented a large modification in its epidemiologic behaviour. Detected in several Brazilian states at the end of the 1980s, the urbanization is the main of them and another is the geographical expansion, associated to a epidemic behavior. Until 1999 São Paulo State had not registered autóctones cases, as an example of this phenomenon. Since that year 1542 patients acquired LVA in São Paulo State (Amato et al., 2009). Before of this LVA urbanization, is emergent the improvement and availability of validated diagnosis techniques, including the molecular tools which could substitute invasive techniques. By this study we intend assess the following visceral leishmaniasis diagnosis techniques, microscopic examination (Giemsa or Panóptico stained smears), culture (NNN/BHI) and kDNA using peripheral blood and bone marrow aspirate samples as templates. We are looking for a minimally invasive and easily performed representing by the peripheral blood techniques which could appear after the assessment of its sensitivity in relationship with the bone marrow.
