Effects of physical exercise on insulin/IGF-1 signaling pathway in brain of diabetic rats

Abstract

Diabetes Mellitus is a chronic disease that has been associated with memory loss, neurological disorders and Alzheimer's disease (AD). Some studies show the importance of the physical exercise to prevent and minimize various neurological disorders. It is believed that the positive effects of exercise on brain function are mediated by insulin and IGF-1. However, there are few studies investigating the effects of physical training on the insulin and IGF-1 signaling pathway of diabetic animals. The main aim of the present study is to verify the role of swimming training on signaling pathway of insulin and IGF-1 in diabetic rats. Wistar rats were divided in sedentary control (SC), trained control (TC), sedentary diabetic (SD), and trained diabetic (TD). Diabetes was induced by Alloxan (32 mg kg(-1) b.w.). Training program consisted in swimming 5 days/week, 1 h/day, 6 weeks, supporting a load corresponding to 90% of maximal lactate steady state (MLSS). At the end of the training period, rats will be sacrificed and sample blood will be collected for determinations of serum glucose, insulin, GH, and IGF-1. Hippocampus and cerebellum will be extracted to determinate protein content and activity by immunoprecipitation and immunoblotting (IR, IGF-1R, IRS-1, IRS-2, PI3-q, Akt, ERK1/2, GSK-3- ², tau protein, APP and ²-Amyloide protein). All dependent variables will be analyzed by two-way analysis of variance (ANOVA) with significance level of 5%. The Bonferroni test will be used for post hoc comparisons. (AU)
