Influence of n-3 fatty acids from fish oil on lipid metabolism and gene expression in rats with nonalcoholic fatty liver induced by fructose

Abstract

The non-alcoholic fatty liver disease (NAFLD) and nonalcoholic steatohepatitis are growing causes of liver disease and is a concern in Public Health. The effects of polyunsaturated fatty acids omega-3 (n-3), are controversial especially regarding of the appropriate amount to the treatment of these diseases. This project aims to elucidate part of lipid metabolism, especially in the formation of triglycerides (TG), liver fatty acid composition of tissues and susceptibility to lipid peroxidation in rats with fructose-induced NAFLD and verify the changes caused by the consumption of n-3 in different proportions from different source in rats. The workwill consist of an induction period of NAFLD, which will be formed two groups, control group (CG, n= 8 rats) and fructose group (GF, n= 32 rats) with the initial weight approximately 200g. During the 60 days will receive GC AIN-93 diet and GF receive 63% fructose. After the initial 60 days, the animals begin Phase II, in which the animals from the CG will form the control group phase II (CII) and continue with the same diet, animals from the GF will be the fructose group phase II (FII) remaining the same diet; GF animals also form other groups containing different amounts of fish oil along with fructose. The animals will be sacrificed after 12 hours of fasting. The animal weight and the amount of forage consumed shall be measured weekly. After the sacrifice will be collected liver, blood, epididymal and retroperitoneal fat for biochemical and molecular analysis. (AU)
