Effect of fat acids on the differentiation and function of the adipose tissue

Abstract

The growing interest in studying the function of adipose tissue is due not only to concerns about obesity and its metabolic consequences, but also by the recognition that adipocytes integrate a wide range of homeostatic processes. Although there are many studies showing the effects of fatty acids (FA) on systemic metabolism and the consensus that the high consumption of saturated FA is associated with cardiovascular risk and insulin resistance, information about the local effect of different FA (saturated and unsaturated) with an pro or anti-inflammatory effect is still incipient. A better understanding of the effects and mechanisms involved in the action of FA in adipocytes wíll have a direct impact on understanding the issues related to obesity and associated diseases, as well as in the development of new therapies that have as a target intracellular pathways located in adipocytes. This project aims to determine the effect of different FA on adipogenesis and the repercussions of these FA on the functionality of adipocytes, with emphasis on regional differences of the various adipose pads (subcutaneous and visceral). The adipogenesis will be evaluated by differentiation of preadipocytes 3T3-L1 cell line, followed by analysis of expression of adipogenic transcriptional factors and lipid content. The functionality of adipocytes, by studying the metabolic and secretory function of the cells already differentiated, being the secretory response assessed by the expression of adipokines and proteins (inflammatory and of insulin signaling pathway) such as leptin, adiponectin, resistin, PAI-1 , IL-6, MCP-1, IL-10, IL-1beta and TNFalpha, IRS-1, AKT, IRbeta and GLUT4; and the metabolic response, by the tests for glucose uptake, lipogenesis, FA oxidation to CO2 and lipolysis on these adipocytes. Ali studies will also be investigated in primary cultured of adipocytes extracted from visceral and subcutaneous adipose tissue of mice. The following FA will be employed on this study: saturated FA (palmitic acid [PA, C16: 0]), mono unsaturated (palmitoleic acid [PE, C16: 1 n-7 and PE, C16: 1 n-9]), and poly unsaturated fatty acids (docosahexaenoic acid [DHA, C22: 6 n-3] and eicosapentaenoic acid [EPA, C20: 5 n-3]).
