Genetic diversity of KPC-producing Klebsiella pneumoniae isolated from several hospitals of São Paulo State

Abstract

The first isolation of KPC-producing K. pneumoniae in Brazil was in 2005, rapidly disseminating to several hospitals in São Paulo State, as well as other States. The aim of this study is to evaluate the genetic diversity of KPC-producing K. pneumoniae isolates from several hospitals of São Paulo State to detect intra and inter-hospital clonal dissemination. Approximately 100 K. pneumoniae isolates sent to Adolfo Lutz Institute and previously confirmed as KPC-producing K. pneumoniae in our laboratory using susceptibility tests (disk-diffusion and microdilution tests) and PCR will be select and submitted to pulsed-field gel electrophoresis (PFGE) and Multi-Locus Sequence Typing (MLST). PFGE will be done according to the CDC protocol, modified by Gautom (1997), genomic DNA will be digested with XbaI (Chef-III, BioRad) and the restriction profile will be analysed by Bionumerics software, version 5.0, 2007 (Applied Maths, Kortrig, Belgium). For MLST, 7 housekeeping genes (Diancourt et al, 2005) will be used and the allelic and sequences type (STs) will be verified on the site http://pubmist.org/kpneumoniae. (AU)
