Molecular investigation of Klebsiella pneumoniae isolates and plasmids carrying beta-lactamases encoding genes

Abstract

Beta-lactamases -producing enterobacterias are globally spread mainly ESBL (CTX-M) and carbapenem resistant-enterobacterial isolates. The objective of this work was study the genetic and molecular epidemiology of ESBL and carbapenem resistant-Klebsiella isolates from patients of Northeast region of Sao Paulo State and the plasmids involved. Identification and antimicrobial susceptibility testing will be accomplished by using semiautomatic systems and standard methods. Clonal relatedness will established by Pulsed-Field Gel Electrophoresis (PFGE) and also by multilocus sequence typing. The presence of genes encoding carbapenemases and extended spectrum ²-lactamases and characterization of bla genes, genetic environment and plasmids will include PCR and further sequencing, RFLP analyses, S1-PFGE and hybridization. This work will point out the high diversity of available genetic elements harboring bla genes. (AU)
