Abstract
The antimicrobial resistance among clinically important pathogens is a public health challenge worldwide, which exhibits a dynamic trajectory from the environment to hospital settings and vice versa. In this regard, resistance is not static, as pathogenic multidrug-resistant (MDR) bacteria strains can move among human and animals populations, as well as among diferent ecossystems. On the other hand, mobile genetic elements carrying resistance genes can move among pathogens and commensal microbiota, favouring the establishment and endemicity of high-risk MDR clones (clonal complexes). The effective management and treatment of infections caused by MDR bacteria, the epidemiology and control of related infections and the discovery and development of new antimicrobial agents increasingly rely upon genotype information "the resistome", supported by next-generation DNA sequencing methods (NGS). This research project aims to obtain a sequence database that allows bioinformatic analysis of pan-resistoma strains of Klebsiella pneumoniae and Escherichia coli producing beta-lactamases (KPC-2, CTX-M-8, CTX- M-15) endemics in Brazil, isolated from human, veterinary and environment samples. The results will contribute to the creation of a reference sequence platform that may be used in diagnostic, therapeutic and/or epidemiological control, among others; elucidating "a priori" genetic aspects of adaptation, resistance and virulence of endemic clones.
