Abstract

In Brazil, the antimicrobial resistance researches in animal origin bacterial strains, including swine, are rare or bit depth. In this regard, the molecular characterization of plasmids harboring antimicrobial resistance genes as bacterial strains carrying these plasmids will be very important to epidemiology and will cause a great impact in antimicrobial resistance research. Plasmids are mobile genetic elements frequently associated to emergence and spread of antimicrobial resistance. The aim of this study is to contribute to clarify the Extended- spectrum Beta-lactamases (ESBL) origin and dissemination in Brazilian animals through molecular characterization of plasmids harboring blaESBL genes extracted from Escherichia coli, Salmonella enterica and Klebsiella pneumoniae isolated from commercial swine stool and establish the clonal relationship between the bacterial strains by Multilocus Sequence Typing (MLST). The antimicrobial susceptibility profile to beta-lactam antibiotics will determined by disc-diffusion method (Kirby-Bauer) and confirmed by Minimal Inhibitory Concentration determination in resistant strains.The ESBL production will be identified by double-disc synergy test (DDT) and research of blaCTX-M, blaTEM e blaSHV genes by PCR. The plasmids harboring ESBL encoding genes will be extracted and transformed into Escherichia coli TOP10 recipient strain and characterized according incompatibility group and molecular weight. The profile of strains carrying these plasmids will be determined by MLST and deposited in specific database (http://pubmlst.org/databases.shtml). (AU)
