Abstract

In the present work will be done to optimize and standardize the method for quantification of metallothioneins in fish tissues using the technique of High Performance Liquid Chromatography with Fluorescence Detector (HPLC-FD). In a second step will be performed several experiments exposing Tilapia to known concentrations of metals (Cd, Cu, Pb and Cr) and different concentrations of Aquatic Humic Substances (SHA) extracted from water sample collected in Municipal Dam of São José do Rio Preto, during dry and rainy seasons, as well as in samples of tap water. Another objective is to evaluate the use of kinetic consumption of hydrogen peroxide as an indicator of lability or recalcitrance of natural organic matter present in a water resource. The AHS will be extracted using the method proposed by the International Humic Substances Society (IHSS). Another objective is to repeat the experiment with samples of water from the dam, making the characterization of physical and chemical parameters such as the concentration of metals, nitrate, ammonia, total organic carbon, pH, conductivity, total dissolved solids, sulfate and phosphate. Quantification of metallothionein in stock gills or liver will allow verification and comparison with biomarkers of exposure developed for the species to be collected. In all tissue samples of fish, liver and gills, as well as in water exposure experiments will be conducted to quantify the total metal stock using a Spectrophotometer Atomic Absorption Electrothermal Atomization (GFAAS) or Flame (FAAS). For the study of the proposed indicator of labile organic matter are mounted in microcosm experiments with water and different concentrations of ascorbic acid and humic acid, which will represent the first labile organic carbon and the second recalcitrant. Each will receive microcosm known amount of hydrogen peroxide (0.24 mg L-1), with controlled temperature and pH throughout the experiment. Will evaluate the influence of UV radiation in experiments by conducting microcosm exposed to light and others kept in a dark environment. A microcosm control is mounted containing only water and hydrogen peroxide. At intervals to be determined due to the decrease in the concentration of hydrogen peroxide. Quantification of hydrogen peroxide is performed using the photometric method sulfate N, N-diethyl-p-feniletilenodiamina (DPD) developed by Bader et al (1988). Total organic carbon, pH and divalent iron will be quantified using Total Organic Carbon Analyzer, pH meter and the spectrophotometric method of orthophenanthroline. Will be determined using mathematical models and the order of reaction half-lives for kinetics consumption of hydrogen peroxide. The results of this work will enable to characterize the impact of metal species to organisms living in water bodies of the region as well as to evaluate the effects of the accumulation of the metal biochemical level, in terms of biomarker response in fish. This study will evaluate the role of the SHA Rio Preto in the availability of Cd, Cu, Pb and Cr for a species of fish. Through comparison of the data intended to propose an indicator of labile organic matter, which could be used as a tool for studies to evaluate the role of organic matter in aquatic environments in all regions of the world.
