Interaction among hypometabolism, calcium dynamics and neuronal excitability: implications in the degenerative process of Alzheimer's Disease

Abstract

Alzheimer's disease (AD) is characterized as a chronic neurodegenerative disorder that affects the individual in adulthood. The brain neurodegenerative process may remain asymptomatic for a long period of time preceding the decline in learning and memory functions, an early sign of pathological changes in AD. Thus, the main challenge is the identification of cellular changes that allow intervention in order to delay, control or prevent the neurodegenerative events before the install to cognitive impairment. Alterations in brain energy metabolism are pointed out as one of the main signs that precede cognitive decline in AD. Several studies show early impairments of the uptake mechanisms and glucose control in the brain of AD patients. GLUT 3, which is the major neuronal glucose transporter, is lowered into the brain of AD patients and modulates a fundamental signal of the pathology, the hyperphosphorylated tau protein. Based on these findings several questions still remain to be clarified in order to elucidate how the decreases in brain energy metabolism could contribute to the development and/or progression of degenerative changes and neuronal modulation of cognitive processes.
