Comparative analysis of physiological parameters and transcriptional profile of different developmental stages of leaf +1 in sugarcane

Abstract

Sugarcane is one of the most important crops in the world and Brazil is its principal producer. Bioethanol importance has increased investments on the acquisition of sugarcane varieties with higher sucrose content. It is known that when sugarcane is cultivated under high CO2 regime it responds with increased photosynthetic rate, plant height, biomass production, water use efficiency, industrial productivity and induction of genes related to photosynthetic system, thus, there is a genetic potential to increase sugarcane productivity. However, physiological and transcriptional evaluation of the different developmental stages present along leaf blade, highly studied in maize, has not been yet reported for sugarcane. The main objective is to evaluate physiological and transcriptional behavior of different segments (2 cm sections) from leaf +1 from sugarcane through (a) measurements of photosynthetic and fluorescence of chlorophyll a parameters throughout the day (6 am - 6 pm); (b) expression of genes related to sink/source transition in order to evaluate tissue maturity along leaf blade during the day; (c) evaluate sugar composition in each leaf segment e correlate them with photosynthetic and expression of sink/source genes during the day; (d) evaluate N content in each leaf segment e correlate them with photosynthetic and expression of sink/source genes during the day; (e) evaluate transcriptional profile using the RNA-seq technique from contrasting leaf segments along the day to identify those genes related to high photosynthetic rate assemble. Our results will allow the identification of new biotechnological targets that could be used to produce sugarcane varieties with higher agronomic performance. (AU)
