Analysis of variability of Bemisia tabaci from the New World group 2 and importance as vectors of Begomoviruses and Criniviruses

The whiteflies (Bemisia tabaci) are recognized as one of the most important pest of the century, mainly because they are vectors of begomoviruses. They are insects with high biological variability and are classified as a complex of species sub-divided into 11 sub groups containing at least 29 species morphologically indistinguishable. Between these species, only Middle East-Asia Minor 1 (MEAM1), New World 1 (NW1) and New World 2 (NW2) were detected in Brazil so far. The species MEAM1, also known as biotype B, is recognized as the largest of worldwide distribution, while species native to the Americas belonging to the group New World are still poorly studied, mainly because they have been discovered (as in NW2) recently and its genetic variability and epidemiological importance in the transmission of begomovirus is still unknown. In this work, we intend to evaluate the capacity of New World 2 species to transmit begomovirus and the number of haplotypes found in the sampled areas. Whiteflies at different locations in the State of São Paulo will be collected and the mitochondrial gene cytochroma oxidase I (mtCOI) analyzed by RFLP and sequencing for identification of species and variability analysis. The presence of begomovirus directly into the insect will be evaluated by RCA-PCR and will indicate whether the species NW2 is a vector or not of begomovirus. Transmissions of begomovirus and crinivirus by specimens of NW2 will elucidate the importance of this species of whitefly in the transmission of this viruses. (AU)
