Abstract

The proteome is the complete set of proteins encoded by the genome and allows to understand how cells adapt to different environmental conditions and provides important information about the proteins quantities and post-translational modifications.The proteomic studies have become an excellent tool to explore, discover and identify the proteins expressed in response to environmental stress, such as carbon source, temperature and pH, because these parameters affect the production of microbial enzymes. Understanding the mechanisms of production and secretion of proteins can be assisted by proteomics and elucidation of these steps may improve the results obtained in bioprocesses. This project intends to continue several subprojects involving prospecting research line of hydrolytic enzymes from microbial source and the enzymes that are targets of this research are proteases, carbohydrases and lipases. The objective of this project is to investigate, using proteomics techniques, the behavior and the enzymes production of the meso and thermophilic filamentous fungi when exposed to different physical and chemical factors. The biochemical potential study of protein expression of each fungus is evaluated by different factors, such as carbon and nitrogen sources, effect of pH and temperature among others. The analyses will be performed by 1D and 2D gels, and the spots differentially expressed in comparison to standard medium will be removed and held on trypsin digestion. The obtained fragments will be applied in the mass spectrometer to obtain the peptides sequences which will be analyzed in silico. The studies on protein expression in filamentous fungi under different conditions of growth open the possibility of exploiting enzymes not yet identified or detected by conventional techniques. Also is possible to discover the best conditions to optimize the enzymes production, considering the biochemical behavior of each fungus in variations of solid and submerged bioprocesses. Thus, the proteomic analyses allow the detection and identification of different proteins expressed in different conditions of microbial growth, contributing to identification of sets protein. (AU)
