Abstract

Although the clinical application of anabolic androgenic steroids (AAS) is recommended for postmenopausal hormone replacement therapy and treatment of sarcopenia and cachexia associated with AIDS, renal failure, cancer and severe burns, many people in modern society have made use of these drugs by aesthetic reasons. This consumption has increased among women in recent decades. It is widely reported that AAS promotes adverse effects on reproduction. However, little attention is given about the effects promoted by AAS in the morphology of the female reproductive tract, and whether these effects are reversible. No studies have evaluated different doses of synthetic steroids administered to the females and the possibility of reversibility of side effects. Thus, the aim of this work is to evaluate the effect of increasing doses of the steroid nandrolone decanoate (1.87, 3.75, 7.5 and 15.0 mg / kg body weight) in the estrous cycle and ovarian tissue of albino rats, and whether a possible recovery of reproductive damage after cessation of steroid treatment. Each experimental group will be composed of 12 females. The control group will receive mineral oil. The treatments will performed by a single subcutaneous injection daily for 15 consecutive days. After the treatment period, six rats for each group will be euthanized and the other six will be submitted to a recovery period of 30 consecutive days without any treatment. The estrous cycle will be monitored daily. At the end of each evaluation period, the reproductive organs will be collected, weighed and the ovaries prepared for immunohistochemical analysis and Western Blot. Will also be evaluated serum concentrations of FSH, LH, Estradiol, Progesterone, Testosterone, DHT and SHBG. The results allow to compare the effects of dose and time-dependent promoted by nandrolone decanoate in gonadal tissue of rats and contribute to the understanding of the molecular mechanisms involved in hormone-receptor interaction in each of the study periods (post-treatment and post-recovery) .
