Effect of TiF4 on hydroxiapatite and enamel subjected to cariogenic and erosive processes: in vitro and in situ studies

Abstract

The aim of this study is to evaluate the interaction and effect of TiF4, included in different formulations (varnish, dentifrice and mouth rinse), on enamel subjected to cariogenic and erosive/abrasive challenges. This project will be divided into three parts. The first study involves doctoral/PQ/IC projects, in which it will be assessed in vitro (A) the quantity of fluoride release into the water from TiF4 and NaF varnishes with different F concentrations (0.95-2.45% F-) along 12h; (B) the type of compounds formed with synthetic hydroxyapatite after the application of TiF4 e NaF solutions using infra-red spectroscopy; (C) the presence of elements on sound and demineralized bovine and human enamel treated with the fluoride varnishes using scanning electron microscopy and microanalysis; (D) the formation of CaF2 on sound and demineralized bovine and human enamel treated with the fluoride varnishes using the KOH-soluble biopsy; (E) the incorporation of fluoride (FAP) using the acidic ans abrasive biopsy and (F) the loss or gain of mineral as well as the lesion depht alteration after the treatments with TiF4 and NaF followed by de-remineralization of bovine or human enamel with artificial carious lesion, changing the remineralization conditions in situ. The change in mineral content will be analysed using transverse microradiography. The second study involves IC project, in which the effect of TiF4, NaF and SnF2 dentifrices (1500 ppm F) against bovine enamel erosion and abrasion will be compared. The quantity of total, soluble and ionic F, pH and REA values of the experimental dentifrices will be checked. A pH-cycling model will be applied, in which the enamel samples will be exposed to the erosive agent (4x90s/day) and abrasive toothbrushing using the experimental dentifrices (2x15s/day, toothbrushing machine), with intervals of exposition to artificial saliva, for 7 days. Enamel loss will be measured using contact profilometry. Finally, the third study will also involves IC, in which the preventive effect of TiF4, NaF and SnF2 mouth rinses (500 ppm F) against bovine enamel erosion will be compared. For this study, the same pH cycling model from the 2nd study will be applied, however, in this case there will be no abrasion. Instead, the samples will be exposed to the fluoride solutions 2x1 min/day. Enamel loss will be measured using contact profilometry. The data will be submitted to the statistical analysis acording to the experimental model (p<0.05). (AU)
