Effect of leukocytes colostrum mother in immune response of newborn calves

Abstract

The objective of this project is to evaluate the effect of passive transfer of leukocytes from bovine colostrum in systemic immunity in newborn calves from birth to 28 days of their life. Will be used 20 calves divided into two experimental groups. The calves receive colostrum COL + group with fresh cells from their mothers; animals COL - group receive acellular colostrum. The acellular colostrum is obtained from a "pool", which is centrifuged to remove cells and frozen at -80 °C. Calves will be assessed at the following times: before feeding colostrum, 24 hours, 2 days (d), 7d, 14d, 21d and 28d after birth. Clinical examination of newborns will be performed and blood samples will be collected into tubes with EDTA (4mL) and anticoagulant heparin (40mL). The innate immune response will be assessed by tests of phagocytosis and release of free radicals by blood monocytes and polymorphonuclear by stimulating bacterial. Specific immunity will be verified by the cultivation of blood lymphocytes and their production of cytokines interferon-gamma (cell response - Th1) and interleukin-10 (humoral response - Th2) stimulated with inactivated bovine herpesvirus (BoHV). Furthermore, immunophenotyping tests will be performed to determine the proportion of the types of leukocytes and their activation. The results obtained using these variables will be tested using the Kolmogorov-Smirnov normality. If they present parametric distribution will be subjected to analysis of variance and differences assessed by Duncan test (SAMPAIO, 1998). The results that do not show this distribution will be tested using the Mann-Whitney test (SIEGEL, 1975). (AU)
