Impact of antioxidant solutions or laser irradiation on bleached dentin after artificial aging of the adhesive interface

Abstract

The post-treatment of the bleached tooth with high concentration of agents aims to eliminate the residual free radicals in order to make it more receptive to adhesive procedures, and also perform restorations in the same session of bleaching treatment. This study aims to assess in vitro the effect of post-treatment with antioxidants (sodium ascorbate, Camellia sinensis - green tea extract and Punica granatum - Pomegranate extract) or laser irradiation (2.64 µm Er:YAG laser and 980 nm Diode laser) in intracoronary dentin bleached with high concentrated agents in different periods: immediate or after 6 months. The sample will be comprised by 512 fragments of intracoronary bovine dentin (5 x 5 mm), with 224 fragments used in the bond strength test (n=14), 160 in the chemical (EDS) and morphological (SEM) analysis at the tooth surface (n=10) and 128 on morphological analysis (SEM) of the adhesive interface (n=8). The fragments will be included, flattened and divided according to the post-treatment of dentin bleached with 35% hydrogen peroxide in 6 experimental levels and 2 controls: GI - no bleaching and no post- treatment (positive control), GII - only bleached (negative control), GIII- stored in relative humidity for 10 days, GIV - 10% sodium ascorbate solution, GV - aqueous extract of Camellia sinensis (green tea), GVI - aqueous extract of Punica granatum (pomegranate), GVII - Er:YAG (2.64 µm) and GVIII - Diode laser (980 nm). Half of the specimens of each group will be analyzed immediately after the post-bleaching treatments and the other half will be subjected to artificial aging of adhesive interface (12000 thermal cycles + 6 months of water storage) for a former analysis of the following response variables: 1) Shear bond strength of the restorative material (MPa), followed by the failure pattern analysis, 2) Quantitative analysis (%) of the levels of calcium, phosphorus and oxygen of dentin bleached and subjected to post-treatments by means of EDS and 3) Qualitative analysis of the surface and adhesive interface morphology of the bleached surface subjected to the post-treatments by SEM. Data will be subjected to appropriate statistical analysis for each test (p < 0.05). (AU)
