Septic response induced in Aeromonas hydrophila Piaractus mesopotamicus: pro-inflammatory and physiological morphological changes

The pathogenesis of sepsis involves multiple interrelationships in the dynamics between the various components related to the host and the pathogen responsible for high mortality rates in several animal species . Although it is a major cause of mortality in fish , no shortage of literature on the pathophysiology of sepsis and its modulation mechanisms . This project aims to study the profile of different variables on the evolution of the septic response induced by Aeromonas hydrophila in Piaractus mesopotamicus . For this, 170 pacu , P. mesopotamicus , with an average weight of 250.71 ± 26.73 g and total length of 25.27 ± 1.60 cm will be used . Initially be given the LD50 ( LD50 - 96h ) to A. hydrophila in pacu . Sepsis is induced by administration of intracelomic LD50 inoculum corresponding to 30 and after me, 1 , 3 , 6 and 24 hours, ten animals in each group ( saline and challenged control) will be anesthetized for blood collection and evaluation of hematological profile , biochemistry , coagulation , free radicals , serum lysozyme , the respiratory activity of leukocytes , the bactericidal lytic activity of serum hemolytic activity of the complement system and activity of bacterial agglutination and the identification and determination of gene expression of tumor necrosis factor ( TNF - ± ) and interleukin 1 ( IL - 1 ) , by reverse transcription method - polymerase chain reaction ( PCR ) in real time. Then the fish will be sacrificed by anesthetic depth for autopsy and collection of material for histopatolagia . Throughout the experiment the animals will be evaluated for clinical changes and the percentage of relative survival . The results are compared by analysis of variance ( ANOVA ) at 5 % probability and the difference between the averages will be compared by the Tukey test . (AU)
