Abstract
The metabolomics is the newest "omics" platform applied in chemical and biochemical studies of organisms, which enables the detection of specific metabolic changes caused by disease states or by pharmacological and/or therapeutic interventions. Although psychiatric illnesses such as schizophrenia and bipolar disorder have been associated with metabolic changes, metabolomics studies in these populations are still scarce. The aim of this research proposal is to compare the metabolic profiles of individuals with schizophrenia and bipolar disorder with healthy controls, and also to investigate the metabolic profile differences between the animal models of schizophrenia and control animals using Nuclear Magnetic Resonance (NMR), principally 1H NMR, and chemometrics. We shall use human and animal plasma samples for NMR analyses and principal component analysis (PCA) and supervised partial least-squares discriminate analysis (PLS-DA) for NMR data treatment. One hundred (150) subjects with psychiatric disorders (50 with schizophrenia, 50 with bipolar disorder, and 50 crack users) will be compared with 50 healthy subjects (control group). Diagnosis will be performed through clinical interview using the Structured Clinical Interview Semi-DSM-IV (SCID). The analysis of the severity of symptoms will be done by using the following instruments: Positive and Negative Syndrome Scale (PANSS), Hamilton Depression Rating Scale (Ham-D), and Young Mania Rating Scale (YMRS). We expect to identify differences among metabolomic profiles of individuals affected by major psychiatric disorders, namely schizophrenia, bipolar disorder and crack users, and the healthy controls. Also, a thorough analysis of differences between the animal model for schizophrenia and control animals using NMR metabolomics will be done, enabling translational studies for posterior follow-up of pharmacological and/or therapeutic interventions.
