The project aims at extracting, separating, identifying, and quantifying the phenolic compounds from aromatic and non-aromatic rice. To ensure that the phenolic compounds are accurately and precisely determined, multiple metabolomic techniques will be used: (A) Broad-spectrum metabolomic profiling using liquid chromatography coupled to time of flight tandem mass spectrometry (LC-MS/MS Q-TOF), liquid chromatography coupled to triple quadrupole tandem mass spectroscopy (LC-MS/MS QQQ), high pressure liquid chromatography (HPLC), and spectrophotometric analyses (to quantify total phenolic levels); (B) Follow-up targeted metabolomic quantifications of key molecules of interest by LC-MS/MS (QQQ and Q-TOF), zonal capillary electrophoresis (CZE), Nuclear Magnetic Resonance (NMR), thin-layer chromatography, and HPLC; (C) Identification of unknowns using LC-MS/MS Q-TOF and NMR. To supplement LC-MS/MS studies, zonal capillary electrophoresis (CZE) and thin-layer chromatography will be used to analyze profile of phenolic compounds from rice, since these methods provide the advantages of being low-cost and high throughput, allowing us to rapidly screen large numbers of rice samples and prioritize them for LC-MS/MS analyses. The research will be conducted in the OARDC Metabolite Analysis Cluster metabolomics facility, in Wooster/Ohio, EUA and in the Nutrient and Phytochemical Analysis Shared Resource (NPASR) metabolomics facility, in Columbus/Ohio, EUA. The research will be supervised by Dr. Joshua Blakeslee from the Department of Horticulture and Crop Science of The Ohio State University with the collaboration of Dr. Joe Scheerens from the same department and Dr. Ken M. Riedl and Dr. Emmanuel Hatzakis from the Department of Food Science and Technology. (AU)
