Abstract

Research on medicinal plants has been of great importance in the search for new pharmaceuticals. A plant with few studies but with potential pharmaceutical applications is Eugenia pyriformis Cambess, a tree species native to Brazil. There are still no studies of the applications of E. pyriformis using the advanced bioanalytical techniques proposed in this project. Techniques such as 1H NMR metabolomics and high-resolution bioactivity profiling/HPLC-PDA-HRMS-SPE-NMR are powerful tools for identification of antidiabetic compounds in plants and accelerate identification of new bioactive compounds. In this project, E. pyriformis will initially be investigated using PCA analysis of 600 MHz 1H NMR spectra of crude extracts, and this, linked to bioactivities of the extracts will allow me to perform covariance analysis and thus identify the individual components responsible for bioactivity. After that, quadruple high-resolution alpha-amylase/alpha-glucosidase/PTP1B/radical scavenging profiling will be performed - which will allow me to pinpoint HPLC-peaks directly correlated with one or more bioactivities. The structure of the bioactive constituents will subsequently be established using HPLC-PDA-HRMS-SPE-NMR. This internship abroad will be of great importance for complementing the results obtained in the doctorate and will enable access to advanced technologies under the supervision of Professor Dan Staerk that will broaden the knowledge of the PhD student related to the study of bioactive compounds from natural sources.
