In vivo and in vitro effect of the antiinflammatory and antioxidant on dystrophic muscle fibers of Mdx mice

Abstract

The objective of this study is to analyze the effects "in vivo and in vitro" the association of inflammatory Diacerhein with antioxidant Vitamin E on the muscle fibers of dystrophic mdx mice. In the studies "in vivo", mdx mice with 14 days of postnatal life will be divided into 4 groups: (1) treated with saline, (2) treated with the combination of Diacerhein and Vitamin E, (3) treated with Diacerhein and (4) treated with Vitamin E. C57BL/10 mice will be used as control and will not receive any treatment. The diaphragm muscle of all experimental groups will be withdrawn and used for morphological and biochemical. Blood samples will also be removed and subjected to biochemical analysis. Studies "in vitro" will be performed with primary cultures of muscle cells of C57BL/10 and mdx mice. Muscle cells are treated or not with Diacerhein and Vitamin E and subjected to biochemical analysis. The results will be statistically analyzed. (AU)
