Abstract

Innate immune responses are the most profound, generic and highly efficient means of host defense against invading pathogens including viruses. Wild birds act as reservoir for several viruses and thus posing continuous threats to both livestock and human beings. Viruses belonging to three main virus families are often found in wild birds: Orthomyxoviridae, Paramyxoviridae and Coronaviridae, that also infect human and other livestock species. The rationales and mechanisms that enable the wild birds as reservoir-host of viruses of proven or potential significance for human and veterinary health are not yet completely elucidated. To this end, we will initially screen clinical material collected from wide range of wild bird and poultry species using family-wide and high-throughput real time RT-PCR assays. The samples appeared positive, for single or multiple viruses, will be subjected to next generation sequencing (NGS) for both amplification and annotation of their complete genomes. The cellular immune responses will be evaluated in immortalized in vitro avian cell culture model system (DF-1) and their possible significance in disease kinetics will be monitored both in single infection and co-infection experimentationsTaken together, the outcome of this proposal will help recognition of viruses that have significant economic impact on the livestock, especially for the poultry of Brazil (third largest producer of chicken meat). In addition, determination of factors, genomic variations and alternation in immune responses against individual and multiple pathogens would help to not only underpin the nature and breadth of immune responses but also may facilitate the establishment of strategies that help to control spread and transmission of viral diseases in wild birds species especially those that are under extinctions or endangered.
