The mesenchymal stem cells (MSCs) are a sub-population of stromal cells, multipotent, with fibroblast features and characteristic surface markers, adherence to the plastic surface of the culture flasks, and under specific induction conditions, MSC are able to differentiate into bone, cartilage and adipose tissue. The use of MSC has been considered as a potentially promising therapeutic alternative in several preclinical and clinical studies. The great interest in MSC for cell therapy is due to their multiple positive features such as: ease of obtaining, low immunogenicity, thus may be used in allogeneic transplants; besides, they can be obtained from various tissue types. Currently, more than 200 clinical trials involving MSC use in different pathological conditions, ranging from immunomodulation to regeneration and repair of tissue injuries, are being carried out. Although, bone marrow has been identified as the first body's source of MSC, other organs and tissues have been identified as potential sources of these cells, such as umbilical cord, placenta, liver, kidney, and more recently, adipose tissue. Among the key challenges of expansion and collection stem cell technologies are the retention of major cell quality attributes with respect to identity, strength, purity and safety. According to these issues, the aim of this project is to establish and standardize a bioprocess for the mesenchymal stem cells multiplication from adipose tissue, which can be scaled-up in good manufacturing practices conditions, by means of a technology based on microcarrier cell culture in stirred tank bioreactor. It will be also developed a monitoring system for metabolism and cell differentiation based on near infrared spectroscopy combined with multivariate techniques of data analysis. Alongside, cell cultures of MSC derived from adipose tissue will be monitored for genomic integrity throughout the process, from plates to bioreactor. (AU)
