Abstract
Obesity is associated with several comorbidities such as dyslipidemia, hypertriglyceridemia, insulin resistance (IR) and diabetes mellitus type 2. Currently the literature reports that these comorbidities are directly related to oxidative stress and inflammation, situations that trigger an imbalance in production adipokine, or increase of proinflammatory cytokines and decrease adiponectin. This condition can cause renal and metabolic disorders, often associated with increased body weight. The ³-oryzanol, main phytosterol present in grains of rice, has shown potential antiadipogênica action, hypocholesterolemic, anti-inflammatory and antioxidant. Thus the aim of this study is to evaluate, in an animal model, the effects of ³-oryzanol on the activation of Adipo-R1 and Adipo-R2 receptors and their role in improvement of renal function. It will be used male Wistar rats (n = 60), 30 days old, from the Universidade Estadual Paulista (UNESP). Initially the animals are divided randomly into three groups to receive standard diet + water (n = 12 animals), fructose diet + 25% fructose in water (n = 36 animals) or fructose diet with ³-oryzanol + 25% fructose in water (n = 12 animals). After the development of albuminury (around 16 weeks), the animals of the FF group (n = 36) will be again divided to receive fructose diet + 25% fructose in water (n = 12 animals) or fructose diet with ³-oryzanol + 25% fructose in water (n = 12 animals) or a standard diet (n = 12 animals) for 8 weeks, totaling five independent experimental groups. Obesity is characterized by adiposity index and co-morbidities: blood pressure, blood glucose, lipid, hormonal and inflammatory serum profiles. They will be measured plasma levels of ³-oryzanol, total antioxidant capacity. In adipose tissue will be analyzed the inflammatory profile for ELISA. Renal function is evaluated by albuminury. It will analyze the inflammatory status and oxidative stress in renal tissue, transmission electron microscopy and morphological tissue study. (AU)
