Phenotypic and genome-wide association studies of the interaction between Burkholderia ambifaria RZ2MS16, a plant growth promoter rhizobacteria, and the tropical maize

Abstract

Burkholderia rhizobacteria may contribute to growth promotion of different crops. This is due to the ability that some bacterial strains have to produce phytohormones, siderophores and nitrogen fixation among other properties. Such processes increase the ability of some species of Burkholderia to be used as bio-fertlizers as a sustainable alternative to the use of chemical fertilizers. Burkholderia ambifaria, strain RZ2MS16, is a Plant Growth-Promoting Rhizobacteria (PGPR) isolated from guarana and it is able to increase the biomass of important crops including maize, soybean, sugarcane. However, little is known about the molecular mechanisms related to growth promotion ability of this strain as well its molecular interaction with the hosts. In other hand Also, preliminary studies have demonstrated that the symbiosis between nitrogen-fixing bacteria (NFB) and maize may be optimized by a co-inoculation using PGPR. A molecular study of the interaction PGPR-hosts and NFB as Azospirillum brasilense received till now little. Similarly, little is known about the host genetic herdability to traits related to the bacterial symbiosis the response to the nutrients that these microorganisms may offer. So, there is a great need to integrate genetic breeding programs with molecular studies of plant-microbe-interactions. Aiming an holistic view of the system PGPR-host-NFB the objectives of the present research are: i) understanding on the genomic level the mechanisms that allow B. ambifarma RZ2MS16 to act promoting increase maize growth; for this, it will be studied the bacterium genome using the knockout of genes related with plant growth promotion and the host-interaction. By transcriptomic, it will be investigated the interaction among the strain B. ambifaria, A. brasilense and tropical maize hybrids and ii) identification of the genetic control and associated SNPs (single nucleotide polymorphisms) related with the maize interaction and the co-inoculated bacteria. For this, it will be analyzed a complete diallel mating design among maize lines contrasting for nitrogen use efficiency, under low and high nitrogen availability in the soil and with and without bacterial inoculation. The phenotypic data will be evaluated using mixed models equations. For the identification of SNPs in linkage disequilibrium with target traits, molecular markers and phenotypic data will be evaluated by genome-wide association studies (GWAS). (AU)
