Effects of excess and restriction of the macronutrientes intake in rats (Rattus norvegicus) during stages of development

Abstract

In order to evaluate the biological and behavioral effects of macronutrients in the diet of Wistar rats fed ad libitum and 30% restriction diet since birth, will be used 60 litters of rats (mother with 8 puppies) distributed in groups: Control (C) - the animals receive standard diet and water ad libitum (n = 6); Protein (P) - the animals receive diet supplemented with 30% of calories from protein and water ad libitum (n = 6); Carbohydrate (CH) - the animals receive diet supplemented with 30% of calories from simple carbohydrates (sugar) and water ad libitum (n = 6); Lipid (L) - animals receive diet supplemented with 30% of calories from unsaturated lipids (lard) and water ad libitum (n = 6); Carbohydrate and Lipid (CHL) - the animals receive diet supplemented with 30% of calories from simple carbohydrates (sugar) and unsaturated lipids (lard) and water ad libitum (n = 6); Restriction (R) - animals receive 70% of the feed intake for C and water ad libitum (n = 6); Protein Restriction (PR) - the animals receive 70% of the diet supplemented with 30% of calories from protein ingested by P and water ad libitum (n = 6); Carbohydrate Restriction (CHR) - the animals receive 70% of the diet supplemented with 30% of calories from simple carbohydrates (sugar) ingested by CH and water ad libitum (n = 6); Lipid Restriction (LR) - the animals receive 70% of the diet supplemented with 30% of calories from unsaturated lipids (lard) ingested by L and water ad libitum (n = 6); Carbohydrate and Lipid Restriction (CHLR) - the animals receive 70% of the diet supplemented with 30% of calories from simple carbohydrates (sugar) and unsaturated lipids (lard) ingested by CHL and water ad libitum (n = 6). After weaning the pups continue to receive the same treatment of mothers and will be evaluated at 70 and 180 days old. They will carry out the nutritional assessment (body weight, naso-anal length, feed intake, weight gain, body mass index, Lee index, coefficient of feeding efficiency), organs weight (spleen, heart, liver, kidneys, adrenals and testes) and abdominal fat (epididymal, retroperitoneal and visceral), blood biochemistry (glucose, triglyceride, total cholesterol and fractions), biochemical (triacylglycerol and total cholesterol) and lipids of faeces and liver, length, total mineral content and calcium from the bones (femur and tibia) and behavioral assessment (Morris water maze, elevated plus maze, object recognition memory related to the specific location and context memory and recognition of two-dimensional stimuli). Data are analyzed by ANOVA followed by Newman-Keuls test (p <0.05). (AU)
