Abstract

The aim of this project is to identify molecular and physiological mechanisms that govern embryogenesis in Brazilian pine, and with that knowledge overcome the recalcitrance to complete the in vitro embryogenic program. To do that, we will combine information from Brazilian Pine embryogenesis and from cell lines of Medicago truncatula with different degrees of embryogenicity. For the identification of the molecular and physiological mechanisms controlling embryogenesis in both systems, we will use proteomic data (2D gel electrophoresis - LC-MS/MS and GeLC-MS/MS based label free proteomic profiling) associated to pharmacological manipulation of endogenous nitric oxide (NO) and polyamine (PAs) levels. We will give particular attention to the post-translational regulation of proteins mediated by S-nitrosylation during in vivo and in vitro embryogenesis. The results will bring new insights on basic and applied plant cell biology, namely in the protein signaling pathways that are regulated by S-nitrosylation and its role in PAs homeostasis in Brazilian pine and M. truncatula, as well as in the ability to use somatic embryogenesis to propagate Brazilian pine. The complementary skills and experience of the Project Proponent, BIOCEL (IB/USP) and BCV-ITQB (Universidade Nova de Lisboa) research teams will be essential to obtain the desired aim. (AU)
