Abstract

Cancer is a complex tissue composed by malignant neoplastic cells surrounded by reactive stroma that contains fibroblasts, as well as inflammatory and endothelial cells, and whose dynamics is currently recognized as primordial for tumor evolution, grown and dissemination. It is believed that almost all solid tumors have atypical fibroblasts in their stromal compartment, known as cancer-associated fibroblasts (CAFs). Proteases secreted by CAFs are frequently positively regulated in tumoral microenvironment and influence neoplastic behavior through tissue architecture breakdown and signaling interactions. In dogs, mast cell tumor is one of the most frequent skin neoplasm and fibroblast are integral components of the connective tissue. These cells synthesize extracellular matrix components, including collagen, fibronectin and laminin, abundant in cutaneous microenvironment. The aim of this research project is to identify, isolate and characterize CAFs in canine mast cell tumors, and investigate their role in maintenance of tumors as dynamic entities and in the promotion of cancer invasive and metastatic potentials.
