Abstract

Diabetes mellitus (DM) is a metabolic disorder that affects 5% of people around the world. Currently, many drugs are used to treat diabetes, including exogenous insulin and allopathic drugs such as biguanidas, sulfonilureias and alpha-glycosidases inhibitors. However, they have many adverse side effects that diminish the quality of life of the patient and the answer for prolonged use. In recent years there has been an increase of research involving medicinal plants in the treatment of many diseases, including DM. However, there are few data to elucidate the mechanism of action of these plants. The genus Bauhinia L. (Fabaceae) consists of approximately 300 species called popularly as "pata de vaca" and have been used in folk medicine for treating diabetes, infections, pain and inflammation. The purpose of this study is to ascertain the mechanism of action of a molecule isolated from the extract of leaves of B. holophylla, a non-cyanogenic cyanoglicoside, named lithospermoside, in streptozotocin-induced diabetic mice. A group of control animals (CTL) and three groups of diabetic animals (STZSAL, STZMET, STZLITO) will receive treatment for 28 days by gavage. CTL and STZSAL groups will receive saline solution. The STZMET group will receive metfomin (300 mg/kg b.w.) and the STZLITO group will receive the lithospermosede (10 mg/kg b.w.). In addition to the general characterization of the effects, through measurements of blood glucose, plasma levels of lipids, ipGTT and ipITT, will be held a proteomics analysis of the liver, with the identification of proteins which are altered by the treatment. Individual genes and protein expressions will be made for more direct access to the mechanism of action. Still, we will analyze the inhibitory activity on ±-amylase and ±-glicosidade enzymes throughout in vitro and in vivo tests. (AU)
