Abstract
Aquaculture is seen as a strategic market for sustainable development, food production and expansion of unexplored frontiers in Brazil. However, the health aspects of production and the lack of infrastructure for the diagnosis of major infectious diseases are considered highly relevant barriers to increase production and productivity of herds in the different regions. The family Iridoviridae, especially ranaviruses, has been responsible for epidemics of great ecological and economic impacts on fish and amphibians of importance in aquaculture from various parts of the world, including Brazil. The diagnostic methods currently available, although they are very effective in detection of ranaviruses, require specialized training and equipment for proper performance and the results are not immediate. Thus, the aim of this project is to develop a direct, sensitive and specific immunochromatographic test for rapid and qualitative detection of ranavirus in samples of fish and amphibians. Therefore, the methodology will consist of three stages, consisting of interdependent experiments: the first experiment will comprise the gene expression of MCP protein of ranavirus (FV3-like); the second experiment will comprise the production of anti-MCP polyclonal antibody in rabbits; and the third experiment will comprise the assembly and validation of the immunochromatographic dipstick test for ranavirus detection. It is expected that a rapid, sensitive and feasible test diagnostic for field conditions can collaborate for a better understanding of the movement of such infectious agents in fish and amphibians from Brazil and thus may promote measures for their control and prevention of ranavirus infection.
