Abstract

Human antimicrobial peptides as LL37 presents a broad spectrum of bactericidal activity against to Gram-positive and Gram-negative bacteria, fungi and enveloped viruses, thus, represent an important defense mechanism against the pathogenic microorganisms invasion. They appear to be engaged in the regulation of both innate and adaptive immune functions in oral cavity. Our aims are to evaluate of LL37 levels in the gingival tissue and in the neutrophils culture isolated from peripheral blood from volunteers with non-inflamed and inflamed gingival. The levels of LL37cathelicidin in gingival tissue will be evaluated by ELISA method using a synthetic peptide and specific policlonal antibodies. Additionally, to study the bacterial interference with the LL37expression and production by neutrophils, these cells will be stimulated with a several bacteria and by some virulence factors. The supernatant of cultures will be submitted to ELISA method to LL37quantification and the cellular mRNA extracted will be used to quantification of cathelicidin through the reaction chain of polymerize in real time (Real Time-PCR) technique.
