Abstract

The aim of this study is to evaluate the effect of sodium hypochlorite on the structural characteristics and viability of microbial cells in biofilms formed on dentin according to the concentration of sodium hypochlorite (1% vs 2.5%), exposure time (5, 15 and 30 minutes), use of chelating solutions, intracanal dressings, root canal dressings, ultrasound or lasers. Sterile bovine dentin blocks will be mounted on a palatal intraoral appliance, which will be used by a volunteer for a period from 72 to 96 hours for the formation of oral biofilm in situ. Then, the dentin blocks will be carefully removed from and washed with 100 mL of PBS. The blocks will be experimentally treated with sodium hypochlorite using different concentrations and times. After the experiment, 500¼l of sodium thiosulfate was used to eliminate waste and inactivate the sodium hypochlorite solution. The samples will be stained with 50 ul of a solution of propidium iodide/Syto9, Live / Dead technique (marker of cell membrane integrity) for 10 minutes for the subsequent analysis of the vitality by using a Confocal Laser Scanning microscope.For the experiments that include the use of ultrasound or lasers will be used transparent acrylic teeth with artificial canal. An external cavity which communicates with the artificial root canal will position the block of infected dentin to simulate the conditions after the intracanal use of ultrasound.Statistical analysis of the values obtained for each group will be calculated by using the "GraphPad Prism" software.
