Abstract

In this work will be made optimization and standardization of the method for quantification of metallothionein in fish tissues using the technique of High Performance Liquid Chromatography with Fluorescence Detector (HPLC-FD). In a second step will be performed several experiments exposing tilapia known concentrations of metals (Cd, Cu, Pb and Cr) and different concentrations of Aquatic Humic Substances (SHA) extracted from water sample collected in Dam Municipal de São José do Rio Preto, referring the dry and rainy season, as well as in samples of tap water. The SHA will be extracted using the method proposed by the International Humic Substances Society (IHSS) and characterization will be performed by UV / Vis and Fluorescence Molecular. Another objective is to repeat the experiment with samples of water from the dam, making the characterization of physico-chemical parameters such as the concentration of Cd, Cu, Pb, Cr, nitrate, ammonia, total organic carbon, pH, conductivity, sulfate and phosphorus . The quantification of metallothionein in tissues of fish allow verification and comparison with exposure biomarkers developed for the species collected by another student in the research group. In all samples of fish tissue and water in the exposure experiments will be carried out quantification of total metal employing a spectrophotometer Electrothermal Atomization Atomic Absorption (GFAAS) or flame (FAAS). The results obtained in this study will enable to characterize the impact of metals to organisms living in water bodies in the region, as well as evaluating the effects of accumulation of metal biochemical level, in terms of biomarker response in fish. This study will assess the role of the SHA Rio Preto in the availability of Cd, Cu, Pb and Cr for a fish species, thus generating unprecedented results for the region to which it belongs. (AU)
