Chronic obstructive pulmonary disease (COPD) is now a major disease in terms of global health. Epidemiological data prove the descent of the number of deaths caused by COPD, with an increase of 340% in 20 years. The causes of COPD are not yet fully understood, but evidence points to the importance of the proteinase-antiproteinase theory as one of the main causes. In this sense, it becomes necessary to use experimental models of lung inflammation to search for new drugs in order to study the importance of inhibition of this mechanism in the control of lung injury. The use of extract of CrataBL for pathophysiological experimental research of COPD rests on the theory of liberation and inhibition of proteinases, altering the development of lung injury caused by elastase. This project aims to assess the effect of CrataBL protein as proteinase inhibitor in experimental model of elastase-induced emphysema in the lungs of mice. We will evaluate the following parameters: respiratory mechanics, exhaled nitric oxide, the mean alveolar diameter, the content of collagen and elastic fibers in the airways and lung parenchyma, macrophages and neutrophils in the bronchoalveolar lavage and lung tissue, lipid peroxidation, the expression of metaproteinases 9 and 12; iNOS, eNOS, isoprostane PGF2 alpha, IL-1, IL-8 and TNF alpha in the lung parenchyma.
