Abstract

In endodontic practice, it is necessary to use biomechanics and the physical-chemical preparation of root canals to obtain a good prognosis and a satisfactory marginal sealing. Those objectives are achieved through the use of sealers and biocompatible with low cytotoxicity which do not harm the surrounding tissues and promote tissue repair apical and periapical. Sealers presenting low toxicity and being biocompatibility are critical since they will be in permanent contact with the tissues of the root canal system and periapical and apical regions. In addition, these cements should provide tolerance to tissue sealing and/or isolation with tissue or mineral fibers. However, these cements stimulate the cells of immune system such as neutrophils, macrophages and lymphocytes and they are activated. Once activated macrophages exert their functions, such as phagocytes of foreign particles, antigen presentation, cytokine production, tumor necrosis factor alpha (TNF-±), nitric oxide and other oxygenates microbicides (H2O2). Neutrophils produce chemokines and cytokines that will generate attraction and activation of neutrophils and several other cells involved in inflammatory response. Among the chemokines, CXCL8 (formerly IL-8) is main factor chemoattractant and activator of neutrophils. Thus, this project aims to establish the production of CXCL8 by macrophages and neutrophils stimulated with sealers, since, for tissue repair, it is necessary not to assault them. Furthermore, viability rate will be analyzed. (AU)
